Hermione, being the intelligent lass that she is, probably fully realizes that a human body is not designed to be 4 inches tall, and that the shrinking of the vital organs would result in them not being able to function (especially concerning breathing and oxygen intake.)
By the looks of Crookshank's spit, however, I doubt any scientific inaccuracies of people-shrinking really matter.  For the wonderful ~Ungapants, who, for winning my '  ain in the Polyjuice' contest, requested a piece with the title/theme 'Hermione's Revenge' (possibly as a follow-up to her piece [link]  ) Don't drink anything you don't mix yourself, boys.
